板栗支链淀粉含量的双波长测定方法 被引量:26

摘要 研究了双波长分光光度法测定板果果仁直链淀粉和支链淀粉的含量。对光度测定的最佳条件和样品前处理进行了详细考察。选择支链淀粉的测定波长为550nm和695nm,直链淀粉的测定波长为615nm和475nm。样品测定时应进行脱脂脱糖处理,直链淀粉浓度在5~60μg/mL、支链淀粉浓度在5~140μg/mL时准确度和精密度均较好。 Dual-wavelength spectrophotometry was applied to the determination of amylopectin in Castanea moUissima BL. According to the absorption spectra of the two complexes, the wavelength of I 2-amylose, 615 nm and 475 nm, and of I2-amylopectin, 550 nm and 695 nm, were selected as the measuring wavelength. Both the exactitude and accuracy are very well when the Concentration Range is obeyed in 5 -60μg/mL for amylose and 5 - 140 μg/mL for amylopectin. It should removed glucose and fat in dry powder before detection.
