抗树突状细胞单克隆抗体对大鼠角膜移植术后植片内IFN-γmRNA表达和血清IL-2水平的影响 被引量:1

摘要 目的用抗树突状细胞单克隆抗体(anti-dendriticcell monoclonal antibody,DC-McAb)抑制树突状细胞(den-driticcell,DC),观察其对大鼠角膜移植排斥反应及植片内IFN-γmRNA的表达、血清IL-2水平的影响。方法以Wistar大鼠为供体,SD大鼠为受体,分A:对照组;A0:阳性对照组;B:腹腔给药组;C:结膜下给药组;D:保留受体上皮瓣组。A、A0、B、C组予常规穿透角膜移植术,D组保留受体上皮瓣。每组术前3d及术日,术后第1、第2、第3、第5、第7、第9、第11天给药,A组:腹腔注射灭菌注射用水0.5ml,A0组:腹腔注射小鼠抗大鼠IgG0.1ml+灭菌注射用水0.4ml,B、D组:DC-McAb0.1ml+灭菌注射用水0.4ml,C组:结膜下注射DC-McAb0.05ml。术后裂隙灯显微镜观察排斥反应情况,记录各组角膜发生排斥反应的时间。A、B、C、D组分别于术后第3、第7、第14和第21天采血清,ELISA法测IL-2水平。A、B组术后第7天取角膜植片,RT-PCR法检测IFN-γmRNA的表达情况。结果A组术后(7.63±0.74)d出现排斥反应,A0组(7.50±0.54)d,与A组相比差异无显著性(P>0.05);B组(17.11±1.17)d、C组(13.86±0.69)d、D组(17.88±0.84)d,与A组相比差异有显著性(P<0.01);C组与B、D组相比,差异有显著性(P<0.01);B组与D组相比,差异无显著性(P>0.05)。A组角膜植片IFN-γmRNA可检测到有较高表达,B组的表达量很少或检测不到,两组之间的差异较为明显。正常SD大鼠血清IL-2浓度为(35.18±2.59)pg/ml,移植术后3d略有升高,7d时明显升高,14d时IL-2的水平达高峰,21d时下降,B、C、D组升高幅度与A组相比明显减少(P<0.05),C组与B、D组相比升高幅度较大(P<0.05),B、D两组之间无明显差异(P>0.05)。结论抗树突状细胞单克隆抗体可抑制大鼠角膜移植排斥反应,延长角膜植片存活时间。 Objective To observe the effects of the anti-dendritic cell monoclonal antibody (DC-McAb) on keratoplasty rejection and the expression of IFN-γ mRNA in the rat cornea, the density change of serum IL-2 after penetrating keratoplasty (PKP). Methods The Wistar rat was used as the donor and the SD rat was used as the receptor. The rats were divided into groups A, A0, B, C and D, Routine PKP was done on the rats in groups A, A0, B and C. The receptor's epithelium was reserved in group D. Medicine was given 3 days before surgery, on the day of surgery, and 1, 2, 3, 5, 7, 9 and 11 d after surgery. The groups were given the following treatments:group A, sterilized saline 0.5 ml, abdominal cavity injection; group A0, mouse anti-rat IgG 0.1 ml+saline 0.4 ml, abdominal cavity injection; groups B and D, DC-McAb 0.1 ml+saline 0,4 ml, abdominal cavity injection; group C, DC-McAb 0,05 ml, subconjectiva injection. Rejective reaction was observed with a slit lamp. The corneas were excised at 7 d after surgery. IFN-γ mRNA was detected by RT-PCR in groups A and B. Serum was acquired at 3, 7, 14 and 21 d after surgery in groups A, B, C and D and IL-2 was examined by ELISA. Results The rejective reaction occurred at (7.63±0.74)d after PKP in group A and (7.50±0.54)d in group AO, and the difference was not significant (P〉0.05) but the difference was significant when group A was compared to groups B, (17.11±1.17)d, C, (13.86±0.69)d and D, (17.88±0.84)d (P〈0.01); There was a significant difference when group C was compared to groups B and D (P〈0.01) but the difference between groups B and D was not significant (P〉0.05). High expression of IFN-γ mRNA in the rat cornea at 7 d after PKP could be seen in group A. There was little or no expression in group B. The serum IL-2 density of the normal SD rat was (35.18±2.59)pg/mL There was a slight increase at 3 d, a big increase at 7 d, peaking at 14 d, and decreasing at 21 d.
