油菜遗传转化体系中卡那霉素浓度的筛选及抑菌剂的选择 被引量:6

摘要 利用根癌农杆菌(Agribacterium tume faciens)介导的下胚轴转化方法,将一抗病基因转化甘蓝型油菜。在遗传转化体系的建立过程中进行了卡那霉素(Km)浓度的筛选及抑菌剂的选择,以确定合适的选择压力及适宜的杀菌剂。试验结果表明:选择培养基中附加25 mg/L Km较适宜;抗性苗筛选过程中羧苄青霉素(Cb)对芽苗分化影响较小,且在500 mg/L时能够很好的抑制农杆菌的过度生长。生根培养基中添加头孢霉素(Cef)和Cb对根分化的影响没有明显差异。 The hypocotyls segments from Brassica napus line 01701 were transformed by using Agrobacterium tumefaciens harbouring the resistant gene. During the processs of establishing regeneration system, the concentration of kanamycin and selection of bacteriostat were tested. Result showed 25 mg/L Km added into selected medium was proper and Cb had little effect on shoot regeneration, also 500 mg/L Cb had best effect on inhabiting Agribacterium. In root induced medium, Cef and Cb had no obvious difference on root differentiaition.
