摘要
冠突散囊菌液体深层发酵滤液经浓缩,醇析,透析冻干得多糖粗品.粗多糖经Sevag法脱蛋白,DEAE-52阴离子交换柱层析纯化得到4种多糖.将含量最高的多糖ECP-A经Sephadex G-100葡聚糖凝胶柱层析得到纯化的冠突散囊菌胞外多糖(ECP-A1和ECP-A2),并测定其相对分子质量.用紫外光谱和红外光谱分析鉴定该多糖,紫外光谱分析未见蛋白质(280 nm)与核酸(260 nm)的特征吸收峰,红外光谱揭示ECP-A1具有典型的多糖特征吸收峰,表明ECP-A1和ECP-A2为单一均匀组分的多糖.
Extracellular polysaccharide(ECP) fraction from E. cristatum was obtained with ethanol precipitation, dialyzation against water and freeze-dry methods. After being purified by deprotein with Sevag method, and further purification with DEAE-52 chromatography, four kinds of polysaccharides were obtained. ECP-A1 and ECP-A2 were purified from ECP-A(the main content of ECP) with Sephadex G-100 chromatography and their molecular weight was 8.61×10^4 D and 1.75×10^4 D. With UV spectrum, we found these two polysaccharides have no absorption peaks of protein and nucleic acid at 280 nm and 260 nm. And IR spectrum revealed that ECP-A 1 has the typical characteristic absorption peaks ofpolysaccharides. All results showed that the ECP-A1 and ECP-A2 were identified as homogenous ones.
