摘要
[目的]探究大麻染色体的制片技术。[方法]对大麻染色体制片技术的预处理液、解离时间、染色时间等进行试验研究,并对其进行染色体数目观察。[结果]研究表明:用大麻根尖在常温下用0.003%的8-羟基喹啉溶液预处理2 h或用饱和对二氯苯处理1.5 h,经卡诺固定液固定30 min后,在1∶1的1 mol/L HCl和45%醋酸60℃下恒温水浴中解离30 s,地衣红染液染色8 h压片镜检,能取得良好的制片效果。大麻染色体数目观察表明,大麻染色体数目2n=2x=20。[结论]该研究为大麻细胞学方面的深入研究提供了资料。
[ Objective] The research aimed to discuss the squashing technique of chromosome in Caanabis sativa L. [ Method] The pretreatment solution, the dissociation time and dyeing time in squashing technique of chromosome in C. sativa were studied. And its chromosome number was also observed. [Result] Under room temperature the root tips of C. sativa were pretreated with 0.003 % 8-hydroxyquinoline solution for 2 h or treated with saturated pdiehlombenzene for 1.5 h. After flying with Camoy stationary liquid for 30 min, they were dissociated in constant-temperature water bath of mixing 1 mol/L HCl with 45% acetic acid at 1 : 1 for 30 s at 60 ℃ and stained with orcein dyeing solution for 8 h for squashing and microscopic examination. Tnus good squashing effects could be obtained. The observation on the chromosome number of C. sativa indicated that the chromosome number of C. sativa was 2n =2x =20. [Conclusion] This research provided data for in-depth study on C. sativa from the cytological aspect.
