摘要
[目的]寻找肉牛精子最佳解冻温度和解冻时间。[方法]将54支利木赞公牛细管冻精在不同温度、不同时间下进行解冻,对解冻后的精液品质进行检测。[结果]解冻后精子活率随解冻温度升高(40~75℃)而升高,而畸形率随解冻温度升高而降低,但高温下(90℃)解冻,精子活率显著低于解冻温度为40~75℃(P<0.05),畸形率显著高于解冻温度75℃(P<0.05);低温(40、75℃)下随着解冻时间延长,精子活率增高,但差异不显著;高温(90℃)下解冻,精子活率与解冻时间长短关系更为明显,时间越长,精子活率越低。精子畸形率、存活时间与解冻时间无显著关系。[结论]在生产实践中根据具体情况选用适合的解冻温度和解冻时间,建议使用75℃/3~5s解冻。
Objective Seek for optimum thawed temperature and time of semen in beef cattle.Method Fifty-four frozen bovine semen straws were thawed under different thawing temperature and times to determine the semen quality after thawing.Result With the increasing of thawing temperature(40~75 ℃)and the rate of motile spermatozoa increased obviously,the abnormal spermatozoa rate first increased and then decreased significantly.But thawing at high temperature(90 ℃),the rate of motile spermatozoa was lower than that thawed at 40 ℃~75 ℃ significantly(P〈0.05)and the abnormal spermatozoa rate was higher than that thawed at 75 ℃ significantly(P〈0.05).Under low temperature(40、75 ℃),with the prolonging of thawing time,the rate of motile spermatozoa increased,but the difference was not significantly.Thawing under high temperature(90 ℃),the rate of motile spermatozoa related to thawing time obviously,the longer the time was,the lower the rate of motile spermatozoa was.There is no relation between thawing time and the rate of motile spermatozoa.Conclusion The suitable thawing temperature and times should be selected according to the practice instance in production and it was suggested to be thawed at 75 ℃ for 3~5 seconds.
