重组人骨形成蛋白2和转化生长因子β复合促进骨生长 被引量:4

Experimenial study on the enhancement of bone formation by recombinant human BMP and TGFβ

摘要 将重组人骨形成蛋白2(rhBMP2)和转化生长因子β(TGFβ)复合后,对比观察其骨诱导及骨生成过程和促进骨生长的效果。以一定比例的TGFβ和rhBMP2与陶瓷化牛骨(CBB)复合,埋入小鼠股部肌肉陷窝内,于第3、5、7、14、21d取材后光镜观察新骨生长情况并计算新骨形成面积。结果发现,rhBMP2和TGFβ复合后其成骨作用比单独应用rhBMP2明显,骨组织较为成熟,新生骨面积14d时2倍于rhBMP2组。而单独用TGFβ无异位诱导新骨形成的作用,但TGFβ可刺激原有骨的膜内成骨。认为,BMP和TGFβ在骨生长修复中各自发挥着不同的作用,TGFβ可促进BMP的骨诱导过程,BMP/TGFβ/CBB复合物或BMP/TGFβ/生物陶瓷复合物具有高效的骨诱导性,在临床骨缺损及骨折修复中有良好的应用前景。 To observe the ectopic bone formation, the complexes of recombinant human BMP2(rhBMP2)/CBB(ceramic bovine bone), TGBβ/CBB, rhBMP2/TGFβ/CBB were prepared and implanted into the thigh muscle pouches of mice respectively. The histological reactions were observed at the intervals of 3,5,7,14 and 21 days after the implantation. The combination of rhBMP2 and TGFβ showed stronger effects in the stimulation of bone formation than rhBMP alone. The quantity of the new bone produced by rhBMP2/TGFβ/CBB was twice as much as that of rhBMP/CBB. TGFβ alone did not induce the ectopic new bone formation, but initiated femoral intramembranous bone formation. It is concluded that: (1) The combination of BMP and TGFβ can not only induce new bone formatin, but also promote the mature of skeletal tisues; (2) TGFβ is the one which is able to influence and promote the process of BMP bone induction;(3) The complex of BMP/TGFβ/CBB or BMP/TGF β/bioceramics may be a high efficient bone inducing material in clinical bone defect repair in the future.
