摘要 目的:观察胰岛素抵抗对不同性别实验大鼠脂肪肝形成的作用。方法:32只Wistar大鼠雌雄各半,按性别不同随机分为4组,雌性对照组、雄性对照组,每组8只。雌性脂肪肝组、雄性脂肪肝组。以复合因子的造模方法饲喂3w后,形成脂肪肝,测定血清及肝组织脂质、血浆内皮素(ET)、ALT、血浆肿瘤坏死因子(TNF)、空腹血糖(FBS)、胰岛素(FINS)的含量变化,计算肝指数及胰岛素抵抗指数并做石蜡切片,HE染色。结果:雌性脂肪肝组、雄性脂肪肝组大鼠肝脏均有重度脂肪变,肝指数、肝组织匀浆甘油三酯(TG)与总胆固醇(TC)、血清TG与TC、ET、TNF、FBC、FINS、胰岛素抵抗指数(HOMA-IR)和ALT均较两对照组升高(P<0.05或P<0.01),雌性大鼠肝组织匀浆TG、ALT、FINS与HOMA-IR均显著高于雄性组(P<0.05或P<0.01),各组大鼠FBG比较,没有统计学意义(P>0.05)。结论:雌性脂肪肝组、雄性脂肪肝组大鼠发生了脂肪性肝炎和胰岛素抵抗,其中以雌性大鼠更加显著。在脂肪肝发生发展过程中胰岛素抵抗的作用可能存在性别差异。 Objective:To observe the function of insulin resistance for male and female rats in the formation of fatty liver. Methods:The 32 Wistar rats with female and male in half each were randomized into 4 groups according to the sex: female control group, male control group, female fatty liver group, male fatty liver group, with 8 in each. They were fed for 3 weeks to make fatty liver models with the compound factor method. The blood serum was tested and the lipin of liver tissue, plasma endotheliolysin (ET), ALT, tumor necrosis factor (TNF), fasting blood sugar (FBS), and fasting insulin (FINS) were measured to calculate liver index and insulin resistance index. The paraffin section was made and HE dyeing used. Results:There was a serious change of fat of liver in the female and male fatty liver groups. The liver index, triglyceride (TG) of homogenate of liver tissue, total cholesterol (TC), serum TG and TC, ET, TNF, FBS, FINS, insulin resistance index (HOMA-IR), and ALT were all elevated than those of the two control groups (P〈0.05 or P〈0.01). The TG, ALT, FINS and HOMA-IR of homogenate of liver tissue of female rats were obviously higher than those of the male group (P〈0.05 or P〈0.01). The FBS in comparison of all groups was not significant in statistics (P〉 0. 05). Conclusion:The fat hepatitis and insulin resistance appeared in the female and male fatty liver groups, especially the female rats. In the developing process of fatty liver, the function of insulin resistance is possibly different for males and females.
