复方番石榴制剂对HepG2细胞胰岛素抵抗的改善及对α-淀粉酶和α-葡萄糖苷酶抑制作用 被引量:5

摘要 目的观察复方番石榴制剂对Hep G2细胞胰岛素抵抗(IR)的改善作用,及对α-淀粉酶和α-葡萄糖苷酶抑制作用。方法用高浓度胰岛素诱导Hep G2细胞建立胰岛素抵抗细胞模型,用葡萄糖氧化酶法检测复方番石榴制剂对IR-Hep G2细胞葡萄糖的消耗量;分别以可溶性淀粉和4-硝基酚-α-D-吡喃葡萄糖苷(PNPG)为底物测定复方番石榴制剂对α-淀粉酶和α-葡萄糖苷酶活性的抑制率。结果与模型组比较,复方番石榴制剂(62.5,250,500μg·m L^(-1))显著增加了IR-Hep G2细胞对葡萄糖的消耗量(P<0.05,P<0.01)。在实验剂量范围内,复方番石榴制剂对α-淀粉酶和α-葡萄糖苷酶抑制作用均随浓度升高而抑制作用增加,IC50值分别为0.79 mg·m L^(-1)和0.48 mg·m L^(-1)。结论复方番石榴制剂可促进IR-Hep G2细胞对葡萄糖的消耗,改善Hep G2细胞对胰岛素的抵抗状态,并能体外抑制α-淀粉酶和α-葡萄糖苷酶活性。 Objective To investigate the effect of compound Fructus Psidii Guajavae preparation on insulin resistance(IR) in HepG2 ceils and its inhibitory effect on ot-amylase and ct-glucosidase. Methods IR-HepG2 cell model wasinduced by high concentration of insulin, and the glucose consumption in IR-HepG2 ceils affected by compoundFructus Psidii Guajavae preparation was detected by glucose oxidase method. The inhibitory effects of compoundFructus Psidii βGuajavae preparation on the activity of a-amylase and ot-glucosidase were examined by usingP-nitrophenyl-β- galactoside (PNPG) and starch as substrate respectively. Results Compared with the model group,compound Fructus Psidii Guajavae preparation at 62.5, 250, 500 μg·m L-1 significantly increased the glucoseconsumption of IR-HepG2 cells (P 〈 0.05 or P 〈 0.01 ). In the range of experimental doses, the inhibition ofct-amylase and ct-glucosidase was increased with the increase of concentration of compound Fructus Psidii Guajavaepreparation, and the ICso values were 0.79 μg·m -1 and 0.48 mg·mL-1. Conclusion Compound Fructus PsidiiGuajavae preparation can promote the glucose consumption of IR-HepG2 ceils, improve the insulin resistance ofHepG2 ceils, and inhibit the activity of tx-amylase and ct-glucosidase in vitro.
