摘要
根据GenBank上发表的犬瘟热病毒(CDV)核蛋白(N)蛋白基因序列,设计合成了1对寡聚核苷酸引物,提取犬瘟热病毒疫苗株CDV3的RNA为模板,采用RT-PCR扩增病毒的N蛋白基因,并进行产物克隆和序列分析。将CDV3疫苗株N基因与GenBank数据库中的疫苗株Onderstepoort及中国、美国和德国等分离的疫苗株或毒株共14株CDV N基因序列进行同源性分析,发现CDV3 N基因序列与疫苗株Onderstepoort的同源性为97.1%,与其他分离毒株的核苷酸同源性为92.8%~94.0%,其中与中国分离的TN株同源性为93.6%;然而,CDV3 N蛋白与它们的氨基酸同源性却基本都为96.6%~97.3%。这说明虽然犬瘟热弱毒株CDV3在核苷酸水平上与其他疫苗株毒株的差异较大,但是在氨基酸水平上的比较差异较小。
Two pairs of PCR primers were designed according to the Nucleocaspid protein gene sequences of canine distemper virus (CDV) in CrenBank. A 1632 bp fragment including Nucleocaspid (N) protein gene was amplified by RT-PCR with RNA template of CDV3. The PCR product was cloned and then sequenced. The homology between the N genes of CDV3 and vaccine strain Onderstepoort and 12 isolated strains in CrenBank database was analyzed. The result showed that the homology of the nucleotide sequence between the CDV3 and vaccine strains Onderstepoort is 97.1% ,and 92.8% - 94.0% with other isolates. There is 93.6% homology between CDV3 and TN in China. However, 96.6% - 97.3% is of amino acid sequence. Then the result showed our vaccine stain CDV3 is closer in zmino acid level than in nucleotide level with other strains.
