加参方对CHF大鼠肾脏水通道蛋白(AQP)mRNA表达的影响 被引量:11

摘要 [目的]通过观察加参方对充血性心力衰竭(CHF)大鼠肾脏水通道蛋白(AQP)mRNA表达的影响,探讨加参方改善水钠潴留的机制。[方法]采用结扎大鼠冠状动脉前降支法造成心肌梗死后形成慢性CHF模型,随机将CHF大鼠分为加参方组、模型组和假手术组共3组,每组8只。连续给药4周。[结果]加参方能够减少CHF大鼠异常增加的抗利尿激素(AVP)水平,抑制AVP-2R受体的激活,降低CHF大鼠肾脏内髓AQP-2mRNA表达。[结论]加参方通过调控CHF大鼠肾脏水通道蛋白AQP mRNA表达,使管腔液中的水通透性减少,减轻CHF时水钠潴留,促进尿液排泄,降低心脏容量负荷,可能是加参方增强心功能的机制之一。 [Objective] To observe the effect of Jiashen prescription on the expression of AQP mRNA of CHF rats and explore the effect mechanism to improve water-sodium retention. [Methods] CHF(congestive heart failure) after myocardial infarction induced by ligation of anterior descending coronary method in rats was established. They were randomly divided into three groups: Jiashen prescription group, model group and sham operated group. [Results] Jiashen prescription may decrease unusual increasing level of AVP and AQP-2m_RNA expression of CHF rats and inhibit AVP-2R activation. [Conclusion] Jiashen prescription may regulate AQP- 2mRNA expression of CHF rats, decrease water permeability of luminal liquid and water-sodium retention, promote evacuation of urine and cut down the volume load of heart. The effect mechanism of Jiashen prescription is probably related to strengthening heart function.
