摘要
采用水酶法提取甜杏仁油和水解蛋白。经筛选,确定选用复合植物水解酶、果胶酶12∶配比后与Alcalase蛋白酶联合作用。通过单因素试验和正交试验,确定了最佳工艺参数:固液比为15∶(W/W),复合植物水解酶与果胶酶添加总量为2.5%,反应时间5 h;碱提pH 8.5,温度60℃,碱提时间60 min;Alcalase蛋白酶反应初始pH 10,反应温度60℃,添加量1.5%,反应时间4 h。在最佳工艺条件下,游离油得率为72.58%,水解蛋白得率为82.35%。
The sugary almond oil and protein hydrolysate were produced using enzymatic hydrolysis method. Complex of cellulase,pectinase (1:2) and Alcalase showed better effect than other methods. The optimal parameters for hydrolysis were established by the single factor and orthogonal test. The optimal processing conditions were as follows: weight ratio of seeds to water 1:5(W/W), the amount of cellulase and pectinase complex 2.5%(V/W),reaction time 5 h;alkaline extraction pH 8.5,time 60 min,temperature 60 ℃ ;Alcalase treatment performed at pH 10,tempreture 60 ℃ with 1.5% (V/W) enzyme for 4 h. Under these conditions, the free oil and protein hydrolysate yield were 72.58% and 82.35%, respectively.
