p21^(WAF1)基因遗传多态性与乳腺癌发病关系的风险 被引量:3

摘要 目的探讨p21WAF1基因多态性与乳腺癌遗传易感性之间的关系。方法应用聚合酶链反应-限制性片断长度多态性(PCR-RFLP)方法检测117例乳腺癌患者和123例健康对照组外周血DNAp21WAF1基因第2外显子密码子31多态性的分布情况。结果乳腺癌与健康对照组间p21WAF1基因第2外显子密码子31的A(arginine)、C(serine)等位基因频率分别54.27%、45.73%及50.00%、50.00%(P>0.05)。携带Ser/Arg和Arg/Arg基因型的个体与携带Ser/Ser基因型的个体比较,乳腺癌患病风险无明显增加(OR,1.108;95%CI,0.579-2.122)。结论p21WAF1基因第2外显子密码子31多态性可能与贵州地区汉族妇女乳腺癌的发病风险无相关性。 Objective To investigate the association of p21^WAF1 gene polymorphism with the susceptibility to breast cancer in Guizhou women. Methods The single nucleotide polymorphisms (SNPS) in codon 31 of p21^WAF1 gene were analyzed by polymorphism chain reaction-restriction fragment length polymorphism(PCR-RFLP) in 117 breast cancer patients and 123 healthy controls. Results The frequencies of A(arginine) and C(serine) allelotype in breast cancer patients and healthy controls were 54. 27 % vs. 45.73 % and 50. 00% vs. 50. 00%(P〉0. 05). The patients with Ser/Arg and Arg/Arg genotypes were at a nonsignificant 1. 108-fold increased risk of breast cancer compared to the individuals with Ser/Ser genotype (95% CI, 0. 579- 2.12). Conclusion In Guizhou women p21^WAF1 gene codon 31 polymorphism may not be associated with development of breast cancer.
