干眼患者泪液和正常人眼角膜、结膜P物质的表达及意义 被引量:4

摘要 目的对干眼患者和正常人泪液中P物质进行测定,研究它在泪液和眼表中的作用。方法应用酶联免疫吸附实验(ELISA)检测非干燥综合征的干眼患者(NSS)17例、干燥综合征的干眼患者(SS)13例及正常人(对照组)28例泪液中P物质的质量浓度,并应用免疫组织化学染色对角膜和结膜组织进行P物质的检测。结果在不同组的所有个体样本中,均检测到P物质的表达,对照组质量浓度为(345.54±18.84)pg/mL,NSS组质量浓度为(260.46±15.87)pg/mL,SS组质量浓度为(173.16±20.58)pg/mL。应用免疫组织化学染色证实人结膜与角膜有P物质的表达。结论SS组和NSS组泪液中P物质的质量浓度均明显降低,和对照组相比差异有统计学意义,免疫组织化学染色证实人结膜与角膜均有P物质的表达。提示P物质对眼表可能发挥营养并具有稳定眼表的作用。 Objective The stability of tear film is a basis to maintain the healthy of the normal ocular surface. Dry eye is caused by the abnormal tear film in the ocular surface. Research showed that aberrant of substance P may be associated with dry eye. We detected the expression of substance P in tears of dry eye patients and normal persons for assessment of its effects in tears and eye surface. Methods The concentrations of substance P in tears were measured by ELISA from 17 non-Sjogren syndrome (NSS) paitents, 13 Sjogren syndrome(SS) patients and 28 normal individuals ( control group). The expression of substance P in human cornea (n = 6) and conjunctiva( n = 6) from the eye bank of Hebei province was analyzed by immunohistochemistry. Results Substance P was detected in all patients. The level of substance P in control group was 345.54 ± 18.84 pg/mL,and that in non-Sjogren syndrome (NSS) group and Sjogren syndrome (SS) group was 260.46 ± 15.87 pg/mL and 173. 16± 20. 58 pg/mL, respectively with statistically significant difference in comparison with control group ( F = 12.31, P 〈 0.01 ). The level of substance P in SS group was significant lower than NSS group ( P 〈 0.05 ). Substance P was expressed in human normal conjunctiva and cornea epithelial cells. Conclusion The level of substance P in SS patients and NSS patients is decreased. Substance P plays important roles in nitrating and stabling ocular surface.
