以50、100、200、400μg/mL的蕨麻多糖对小鼠脾淋巴细胞在体外分别孵育4、8、12、24 h,测定了细胞培养液中一氧化氮(NO)、6-酮-PGF1α及血栓素B2(TXB2)的水平和细胞内cGMP的含量,测定了加入蕨麻多糖后2、5、10 min时细胞内游离钙离子(Ca2+)的浓度。结果显示,蕨麻多糖能明显促进小鼠脾淋巴细胞分泌一氧化氮,显著升高小鼠脾淋巴细胞内cGMP及钙离子的水平,对细胞内TXB2的产生有抑制作用,能够调整花生四烯酸体系向6-酮-PGF1α一端偏移。提示蕨麻多糖能够增强脾淋巴细胞的免疫作用。
Murine la anserine polysaccha splenic lymphocytes were incubated in medium alone or medium containing Potentilride(PAP) in the concentrations of 50 μg/mL,100 μg/mL,200 μg/mL or 400 μg/mL for 4 h,8 h, 12 h or 24 h in vitro, respectively. The supernatants at different culture periods were analyzed for changes in levels of nitric oxide(NO), 6-keto-prostaglandin F1α and thromboxane B2 (TXB2). The cells were collected to determine contents of cGMP. The changes in levels of the concentrations of cytoplasmic free Ca^2+ were measured at minute 2,5 and 10 post-incubation, respectively. PAP increased the production of NO and the concentration of intracellular free calcium in splenic lymphocytes and raised the levels of cGMP and 6-keto-PGF1α of murine splenic lymphocytes in vitro. The levels of TXB2 dropped in the cells treated with PAP. PAP was able to regulate arachidonic acid signal transduction of immunocytes. These results showed that PAP was able to enhance immune functions of the splenic lymphocytes.
