摘要
目的研究大黄酚和大黄素甲醚混合物的高效分离方法。方法采用硅胶柱色谱进行分离,薄层色谱跟踪检测,HPLC检测产品纯度。结果在以工业级柱层析硅胶(100~200目)为填充剂的色谱柱上,以石油醚-乙酸乙酯-甲酸(体积比100∶1∶0.5)为洗脱剂,可以将大黄酚和大黄素甲醚完全分离。结论硅胶柱色谱可以实现大黄酚和大黄素甲醚的高效分离;产品经HPLC检测,纯度达99%以上。
Objective To study the efficient and simple method of separation of chrysophanol and physcion. Methods Using silica gel column chromatography for separation,TLC for tracking and detection, HPLC for purity detection of the products. Results Chrysophanol and physcion could be completely separated by industrial silica gel column chromatography [ 100 ~ 200 mesh, petroleum ether-ethyl acetate-formic acid (100: 1:0.5) as elute 1. Conclusion Chrysophanol and physcion could be effectively separated by silica gel column chromatography. Their purities were ≥99% by HPLC method.
