氧氟沙星等药物联合应用对创伤弧菌感染小鼠的保护作用

摘要 目的探讨氧氟沙星等药物联合治疗对创伤弧菌(Vv)感染小鼠的保护作用及对小鼠肝细胞超微结构的影响。方法清洁级ICR小鼠80只,随机分4组:阴性对照组10只,阳性组25只,氧氟沙星单独治疗组20只,氧氟沙星与其他药物联合治疗组25只。阴性对照组腹腔注射0.9%氯化钠注射液0.2ml,阳性组与药物治疗组腹腔注射Vv增菌液0.2ml(约107CFU/ml),1h后单独治疗组注射氧氟沙星,联合治疗组注射氧氟沙星、肝素钠、地塞米松、异丙嗪、间羟胺和多巴胺。比较阳性组和治疗组小鼠腹腔注射Vv后12h存活率及小鼠肝组织各时段超微结构的动态改变。结果12h后,阴性对照组存活率100%;阳性组小鼠全部死亡,存活率为0;单独治疗组小鼠存活10只,存活率50%;联合治疗组小鼠存活24只,存活率96%。联合治疗组小鼠存活率与阳性组及单独治疗组相比差异均具有统计学意义(均P<0.01)。经电镜观察,阳性组3h后即出现肝内皮细胞扩张、肿胀,细胞核固缩,线粒体肿胀、嵴模糊或消失;5h肝细胞核进一步固缩,线粒体空泡样变性,肝细胞内质网空泡样改变。单独治疗组3h、5h时肝细胞存在不同程度的线粒体肿胀,内质网空泡样改变,并见核固缩。联合治疗组肝细胞病变轻于阳性组及单独治疗组。结论氧氟沙星等药物联合治疗在一定程度上能改善Vv所致的肝细胞进行性损伤,对Vv感染小鼠有一定的保护作用。 Objective To investigate effects Ofloxacin on the liver uhrastructure changes of mice infected by Vibrio vulnificus. Methods Eighty ICR mice were divided into 4 groups: negative control (n=10), positive control(n=25), ofloxacin treatment (n=20), ofloxacin combination treatment(n=25).Mice in negative control group were intraperitioneally injected with 0.2ml physiological saline; Mice in other three groups were injected with 0.2ml (107CFU/ml)bacteria suspension i.p., corresponding medicine were given in treatment groups lh after infection. Compare the liver ultra structure changes of combination groups with the other groups, at the same time, the survival rates of each groups were also compared. Results After 12h the survival rates of the four groups were 100%, 0, 50%, 96% (P〈0.01). Swell of endothdial cells, nucleus pyknosis, the appearance of swollen mitochondria the disappearance of the crest and vacuoles of liver cells were observed in the positive control group, however, these pathological changes were ameliorated in combination treatment group. Conclusion Ofloxacin combined with other agents may effectively protect liver from the damage by Vv infection.
