摘要
目的检测不同口腔鳞癌细胞的缺氧诱导因子-1α的表达水平,探讨口腔鳞癌细胞对顺铂的敏感性是否与其表达水平相关。方法将OSC2、OSC4、OSC5、OSC6口腔鳞癌细胞培养并检测其缺氧诱导因子-1α的表达水平,利用四唑蓝比色法(MTT)检测各种细胞对不同浓度顺铂的生长抑制率。结果OSC2和OSC4的缺氧诱导因子-1α的表达水平明显低于OSC5和OSC6,OSC2和OSC4对顺铂的敏感性高于OSC5和OSC6,其生长受顺铂抑制明显。结论缺氧诱导因子-1α的表达水平与口腔鳞癌细胞对顺铂的敏感性相关,缺氧诱导因子可能成为肿瘤化疗药物的新分子靶点。
Objective To examine the relationship between expression of hypoxia - inducible factor - 1 alpha ( HIF -1α) and sensitivity of oral squamous cells to cisplatin chemotherapy, Methods OSC2, OSCA, OSC5 and OSC6 cells were cultured and expression of HIF-1α was respecdvely examined. Inhibition rate to cisplation was respectively examined by MTT assay, Results Compared with OSC5 and OSC6, the expression of HIF-1α was lower in OSC2 and OSCA. OSC2 and OSCA were obviously inhibited by cisplatin, Conclusion Sensitivity to cisplatin is associated with the expression of HIF - 1. HIF -1α may be potential molecular target for chemotherapy drugs,
