摘要
通过将F904对人外用血单个核细胞(PBMC)体外作用的实验研究,对F904的免疫抑制作用及其机制进行了初步的探讨。结果表明:(1)F904能显著抑制植物血凝素(PHA)诱导的T细胞增殖反应;(2)F904对PHA激活的T细胞表面IL-2R的表达里强烈的抑制作用(P<0.0001);(3)增加培养细胞孔中的IL-2的含量可在一定程度上逆转由F904所致增殖抑制作用,本研究提示:降低IL-2R的表达和抑制IL-2的分泌很可能是F904免疫抑制增殖作用的基础。
The in vitro effects of F904 on human single peripheral blood monocyte (PBMC) were experimentally studied to evaluate the immunosuppressive effect of F904 and its mechanism.The results showed: (1)F904 exerted marked inhibitory effect on PHA-induced T cell proliferation,(2)F904 strongly inhibited the expression of IL-2R on the surface of T cells activated by PHA (P<0, 0001 ), and (3)adding exogenous IL-2 to F904 treated lymphocytes resulted in the reversal of theantoproliferative effect to a different extent. The results suggest that the inhibitory effect of F904on T cell proliferation is most probably mediated by blocking IL-2 production as well as decreasingIL-2R expression.
