摘要
目的观察β-淀粉样蛋白1-40(Aβ1-40)大鼠海马双侧注射后大脑组织Bax和Bcl-2 mRNA表达的变化及加减地黄饮子的干预作用。方法采用大鼠海马立体定向注射凝聚态Aβ1-40诱导老年性痴呆(Alzheimer’s disease,AD)动物模型。采用实时荧光定量PCR法检测大鼠大脑组织Bcl-2和Bax mRNA的表达。结果与假手术组比较,模型组Bax2-ΔΔCt显著升高,而Bcl-22-ΔΔCt显著降低,差异具有统计学意义(P<0.05);与模型组比较,盐酸多奈哌齐组、加减地黄饮子组Bax2-ΔΔCt显著降低(P<0.05),而加减地黄饮子组Bcl-22-ΔΔCt显著升高(P<0.05)。结论海马注射Aβ1-40可诱导大鼠脑组织BaxmRNA表达上调和Bcl-2表达下调,而加减地黄饮子可通过抑制海马组织BaxmRNA的表达和促进Bcl-2mRNA表达,进而起到减少神经细胞凋亡的作用。
Objective To observe the effect of modified "Rehmanniae Decoction" on Bax and Bcl-2 mRNA expressions in the brain tissue in amyloid-1β1-40 induced AD rats in vivo. Methods AD model in rats was induced by amyloid-β1-40 sterotaxis injection into bilateral hippocampus. Modified "Rehmanniae Decoction" was given by oral administration for 28 days. All rats were sacrificed on 5th week. Bax and Bel-2 mRNA expressions in brain tissue were assayed by real-time quantitative reverse transcription-PCR. Results The Bax mRNA expression was upregulated in model groups compared with the sham group and Bcl-2 was down-regulated. In modified "Rehmanniae Decoction" group, the Bax mRNA expression was down-regulated and Bcl-2 up-regulated compared with the model group (P〈0.05). Conclusion Amyloid-β1-40 injection into the bilateral hippocampus can up-regulate expression of Bax mRNA and down-regulate expression of Bcl-2 in brain tissue. Modified "Rehmanniae Decoction" may down-regulate expression of Bax mRNA and up-regulate expression of Bcl-2 mRNA.
