爆炸冲击伤大鼠脑组织c-Fos和c-Jun蛋白表达的改变 被引量:3

CHANGES OF THE EXPRESSION OF c-FOS AND c-JUN IN BRAIN TISSUES AFTER EXPLOSIVE BLAST INJURY IN RATS

摘要 目的探讨爆炸冲击伤对大鼠脑组织c-Fos和c-Jun表达及分布的影响,为进一步阐明颅脑冲击伤的发生机制和指导临床治疗提供实验依据。方法应用爆炸致伤大鼠,采用免疫组织化学法,观察大鼠冲击伤后c-Fos和c-Jun蛋白在脑内的表达及分布情况。结果爆后大鼠脑内各部位均有c-Fos和c-Jun表达,但在时间与空间分布呈不均一性。脑皮层区于爆后12 h c-Fos和c-Jun蛋白均高表达,其阳性细胞表达率分别为(39.3±3.2)%和(37.4±0.1)%;随着爆后时间的延长,其c-Fos和c-Jun蛋白阳性表达率逐渐降低(P<0.01);海马区于爆后4 d c-Fos蛋白阳性细胞表达率高达(29.1±1.9)%,而爆后24 h c-Jun蛋白阳性细胞表达率达到峰值为(42.0±2.2)%;丘脑区于爆后4 d c-Fos和c-Jun蛋白阳性细胞表达率均达高峰。结论爆炸冲击伤脑组织中c-Fos和c-Jun高表达,并参与对神经细胞损伤过程。  Objective To investigate the influence on the expressions of protein c-Fos and protein c-Jun in rat brain tissues after explosive blast injury,in order to provide the experimental basis for further elucidating the mechanism of craniocerebral blast injury,and for clinical therapy.Methods The brain tissue injury in rats was induced by explosive blast.Immunohistochemical method was used to observe the expressions and distributions of protein c-Fos and protein c-Jun in rat brain.Results After explosive blast,expressions of protein c-Fos and protein c-Jun were observed all over the brain,but it was not uniform in space-time distribution.In the brain cortex,high expressions of protein c-Fos and protein c-Jun were observed 12 h after explosive blast,the rates of positive cell expressions of protein c-Fos,and protein c-Jun were(39.3±3.2)%,and(37.4±0.1)% respectively,but the rate of expressions reduced with the passage of time(P〈0.01).In the hippocampus,the peak value of expression rate of protein c-Fos was(29.1±1.9)% 4 d after blast,and that of protein c-Jun(42.0±2.2)% at 24 h.In the thalamencephalon,the rates of expressions of protein c-Fos and protein c-Fos reached peak value 4 d after explosive blast.Conclusion The high expressions of proteins c-Fos and c-Jun are involved in the process of neuron injury in brain tissue after explosive blast damage.
