Determination of Amino Acids in Cerebroprotein Hydrolysates with Pre-column Derivatization by High Performance Liquid Chromatography

摘要 通过升高色谱柱温度,结合线性梯度洗脱的分离模式调整色谱分离的选择性,提出了一种测定脑蛋白水解物注射液中氨基酸含量的2,4-二硝基氟苯(DNFB)柱前衍生高效液相色谱方法。本法采用Kromasil C18(250×4.6mmi.d.,5μm)色谱柱,柱温44℃,在30min内以3个线性梯度能完全分离17种氨基酸,定量线性范围为0.2~1.0mg/L;加标回收率为89%~108%。该法具有适用性好、稳定性高等特点,也可以应用于其它种类样品中氨基酸的测定。 The separation selectivity of amino acids derived with DNFB is regulated with the method of raising temperature of chromatographic column and a linearity gradient program in this study. Based on this, a new method of the determination of amino acids in cerebroprotein hydrolysates by high performance liquid chromatography is established. The amino acids are determined directly under 44℃ with Kromasil C18 (250 × 4. 6 mm i. d. , 5 μm) as chromatographic column. 17 amino acids can be separated primely with 3 linearity gradient in 30 minutes. The quantitative linear range is 0.2- 1.0 mg/ L, and the recoveries for the spiked samples are 89%- 108 %. This method has been applied to the determination of amino acids in cerebroprotein hydrolysates. The developed method is stable and can also be applied to the determination of amino acids in other samples.
