11β-羟化类固醇脱氢酶1型基因微卫星多态性与肥胖的相关性研究 被引量:3

摘要 目的:探讨中国南京汉族人群中11β-羟化类固醇脱氢酶1型基因多态性与肥胖是否存在相关性。方法:选择11β-羟化类固醇脱氢酶1型基因附近高杂合度微卫星DNA多态标记,应用聚合酶链反应及变性聚丙烯酰胺凝胶电泳技术,探查这一微卫星多态在中国南京汉族人群156例(对照组)与128例肥胖症患者(肥胖组)中的基因频率分布差异。结果:该多态位点存在8种等位基因,CA重复序列分别重复13、14、15、16、17、18、19和20次,病例-对照群体分析结果表明,两组间等位基因频率总体分布差异无统计学意义(χ2=7.583,P=0.361)。但CA18等位基因肥胖组频率高于对照组,差异有统计学意义(χ2=5.371,P=0.020),可初步认为CA18等位基因与肥胖发生有关。结论:南京地区汉族人群中11β-羟化类固醇脱氢酶1型基因多态性可能与肥胖有一定关系。 Objective: To explore the relatlonsmp oetween obesity andmicrosatellite DNA polymorphism of human 11 beta-hydroxysteroid dehydrogenases gene 1 ( 11β-HSD1 ) among the Chinese Han population in Nanjing. Methods:A total of 128 patients with obesity and 156 normal subjects were collected ran- domly from the Chinese Hans in Nanjing,. Polymorphism of the microsatellite marker was detected by performing polymerase chain reaction, and the distributions of 11β-HSD1 gene microsatellite (CA)n in the patients with obesity and the controls were compared. Results: Eight types of alleles were identified with different CA-repeat numbers: 13, 14, 15, 16, 17, 18, 19 and20 in the Chinese Hans. The general prevalence of 11β-HSD1 gene alleles in the patients with obesity was not significantly different from that in the controls (X2 =7. 583, P =0. 361 ), though the (CA)is allele differed significantly between the two groups ( X2 = 5. 371, P = 0. 020 ). Conclusion : Polymorphism of microsatellite markers in 11β-HSD1 gene may be associated with obesity in the Chinese Hans in Nanjing.
