摘要
本试验采用可见分光光度法测定芝芪菌质中的总皂苷含量。通过对测定波长的选择,对香荚兰醛浓度、高氯酸用量、加热时间和加热温度的优选,对显色后样品稳定性的考察,确定了芝芪菌质中总皂苷含量测定的最佳条件,并且检测了3批芝芪菌质总皂苷的含量。结果表明:在优选后的最佳含量测定条件下,黄芪甲苷对照品质量在53~185.5μg时有良好的线性关系(r=0.9998);3批芝芪菌质中总皂苷的平均含量为0.2685%。
The experiment determined the content of total saponins in Zhiqi fungal substance by visible spectrophotography.The optimal determination conditions of total saponins in Zhiqi fungal substance was ascertained by studying the wavelength ,the concentration of vanilin,the dosage of perchlonic acid ,the time and temperature of heating,and the stability after coloration.The contents of total saponins of three batches Zhiqi fungal substances were also determined.The results showed there was good linearity (r=-0.9998)on Astragaloside IV during the range of 53 ~ 185.5 txg with the optimal determination condition, and the average content of total saponins of three batches Zhiqi fungal substances was 0.2685 %.
