摘要
建立反相高效液相色谱法测定了山茱萸中乌索酸和齐墩果酸的含量。色谱柱为Kromasil C18柱(250mm×4.6mm,5μm),流动相为甲醇∶水∶磷酸(90:10:0.2),流速0.8ml/min;柱温28℃;检测波长为210nm。乌索酸的线性范围0.67~6.03μg,r=0.9996;齐墩果酸的线性范围0.66~5.94μg,r=0.9996。乌索酸和齐墩果酸平均回收率分别为97.6%和99.8%,RSD分别为1.6%和2.1%。方法简便可行,灵敏、准确,重现性好,可作为控制山茱萸药材质量的检测方法。
A RP-HPLC method for content determination of ursolic acid and oleanolic acid in Comus officinalis was established. The Kromasil C18 (250 mm×4.6 mm, 5 μm) was used as analytical column with methanol-water-phosphate acid (90:10:0.2) as mobile phase. The detection wavelength was set at 210nm, The flow rate was 0.8 ml/min and the column temperature was 28 ℃, The calibration curve of ursolic acid was linear over the range of 0.67-6.03 μg and the correlation coefficient was 0.999 6. The calibration curve of oleanolic acid was linear over the range of 0.66-5.94 μg and the correlation coefficient was 0.999 6. The average recoveries of ursolic acid and oleanolic acid were 97,6 % (RSD= 1,6 %) and 99.8 % (RSD=2.1%), respectively. The method was simple, sensitive, accurate and reproducible. It could be used to control the quality of Comus officinalis.
