海尔福口服液对铅染毒小鼠脑组织O_2~影响的实验研究 被引量:4

摘要 目的:通过海尔福口服液对铅染毒小鼠脑组织超氧气阴离子(O2^-)自由基的影响实验,探讨海尔福口服液的排铅机理。方法:用2g/L,浓度的醋酸铅,按14mg/kg体重剂量,给60只小白鼠作腹腔注射,每天1次,连续15天染毒。然后分成3组;大剂量治疗组、小剂量治疗组、模型组。另设正常组(对照组),用小鼠10只,不染毒。15天后停止染毒,用海尔福口服液按大小二种不同剂量,给大剂量治疗组、小剂量治疗组灌胃连续15天。模型组和正常组用等体积的蒸馏水灌胃,方法和时间与治疗组相同。测定实验前后各组小鼠血红蛋白(Hb)。实验结束,从眼球取血,分离血清,测定谷丙氨酶(ALT)活力和尿素含量。处死动物,迅速取大脑在冷冻下匀浆,测定O2^-浓度和清除率、乙酰胆碱酯酶(TchE)活力、蛋白质。结果:大荆量治疗组、正常组脑组织O2^-清除率明显高于模型组和小剂量治疗组,而大剂量治疗组和正常组脑组织O2^-浓度明显低于模型组和小剂量治疗组(P〈0.05)。小剂量治疗组和正常组血尿Hb明显高于模型组和大剂量治疗组,(P〈0.05)。血清ALT、尿素、脑组织TchE活力、蛋白质各组之间尚无明显差异(P〉0.05)。结论:海尔福口服液有明显清除铅染毒小鼠脑组织O2^-作用。 Objective:To investigate the mechanism of expelling lead of Haierful Oral liquid by the experiment study of Haierful Oral liquid on the effects of cerebral O2^-, in lead poisoned mice. Methods=60 mice were given ip injection by 4 mg/kg( 1 kg mice given 4 mg lead )with 2 g/L of lead aceuc density,once a day until 15 days,then separated them into three groups: treated group 1,treated group2 and lead model group. Plus,there were 10 mice as a control group. After 15 days Lead poisoning,mice of groupl and group 2 were treated for 15 days by Haierful Oral liquid with high dose and low dose which by given it down the stomach,while model group and control group by distilled water. Determined Hb of all animal before and after experiment. And at the end of the experiment, blood from eyeballs was got and isolated, then determined ALT activity and urea content. After the animal were sacrificed, brains was got and homogenized in ice box. then determined the density and clearance of O2^- , TchE activity and protein. Results:Clearance of Cerebral O2^- in group 1 and control group were significantly higher than those in group 2 and model group,but density of the former were lower(P〈0.05). Serum Hb in group 2 and control group were significantly higher than those in groupl and control group,and were significantly higher than those in group 1 and model group(P〈0.05). But still no significant difference of ALT in serum, urea,TchE activity and brain protein among each groups( P)〉 0.05). Conclnsion:Haierful Oral liquid has significant function on eliminating Of out of brain in lead poisoned mice.
