摘要
目的:研究血清与血浆中血栓蛋白的差异,探索临床检验血标本的最佳取材方式。方法:采用表面增强激光解吸/离子化飞行时间质谱(SELDI-TOF-MS)技术,并选用CM10蛋白质芯片对8名志愿者的血清和血浆标本分别进行检测,经Proteinchip Software 3.2软件采集原始数据后,采用Ciphergen公司的Biomarker Wizard软件分析血清及血浆标本中所有质荷比(m/z)蛋白丰度的差异,以初步探讨血清和血浆中血栓蛋白的差异。结果:经Biomarker Wizard软件分析,显示2类标本间共有18个蛋白丰度有显著性差异,其中m/z为1872、1902、1923、1993、2144、2193、2745、3402、15147的9个蛋白在血浆中含量较高,而m/z为3162、4097、4215、5645、5913、7774、8150、8924、9299的9个蛋白在血浆中含量较低。结论:血清和血浆间存在许多血栓蛋白丰度的差异。因此,在临床检验血液中血栓蛋白时,为尽可能反映血液非细胞成分的在体特征,标本选择血浆可能更佳。
Objective To study proteomic differences between serum and plasma for selecting the optimal specimen in proteomic research and laboratory testing.Methods Surface enhanced laser desorption/ionization time-of-flight mass spectrometry(SELDI-TOF-MS) CM10 Proteinchip technology was used to detect eight serum and plasma samples.Raw data was collected by Proteinchip Software 3.2 and then Biomarker Wizard software was used to compare all protein peaks between serum and plasma.Results Eighteen protein peaks were significantly different between the serum and plasma samples.Among them,the concentration of 9 proteins with the key mass/charge(m/z) values 1 872,1 902,1 923,1 993,2 144,2 193,2 745,3 402 and 15 147 were significantly higher in the plasma than that in the serum;while the concentration of 9 proteins with the key m/z values 3 162,4 097,4 215,5 645,5 913,7 774,8 150,8 924 and 9 299 were dramatically lower in the plasma.Conclusions Compared with plasma,there are many significantly different protein peaks in serum samples;plasma should be used to maintain concordance with the body and reflect the real state of cell-free component in blood when performing proteomic studies and laboratory testing.
