摘要
目的比较不同培养条件下马尔尼菲青霉(Pm)菌丝相及酵母相的异柠檬酸裂解酶(ICL)基因转录水平,初步探讨乙醛酸循环在Pm致病中的意义。方法将Pm分为高糖培养的菌丝相(MH)、低糖培养的菌丝相(ML)及高糖培养的酵母相(YH)、低糖培养的酵母相(YL)4组,采用实时定量RT-PCR法比较各组ICL基因转录水平。结果MH、ML、YH、YL组ICL相对表达量分别为1.000、3.000、3.654、69.530,组间差异有统计学意义(P<0.05);温度增高和葡萄糖浓度降低两因素之间具有协同效应。结论Pm酵母相的ICL转录水平较菌丝相增高,而且低糖培养时增高更为显著,提示乙醛酸循环可能在Pm致病过程中起着重要的作用。
The differentiai expression of iso-citrate lysae in mold- and yeast phase of Penicillun rnarneffei was compared in order to explore the significance of the glyoxylate pathway in the pathogenesis of this fungus, in which the transcriptional level of iso-citrate lysae in 4 tested groups of P. marneffei were detected by using real time RT-PCR. It was found that the transcriptional levels of iso-citrate lysae in mold phase in high-sugar culture (MH), mold phase in low-sugar culture (ML), yeast phase in high-sugar culture (YH) and yeast phase in low-sugar culture (YL) groups were 1. 000, 3. 000, 3. 654 and 69. 530 respectively, with statistical inter-group differences (P〈0.05). Synergistic effect existed between increase of temperature and reduction of glucose concentration. Transcriptional level of iso-citrate lysae induced in yeast phase was higher than that in mold phase, and the lower concentration of glucose could urge this process. It is presumed that the glyoxylate pathway may be important in the pathogenesis of P. marneffei.
