摘要
目的:探讨脂源性炎症因子TNF-α,IL-6及IL-18在2型糖尿病大鼠视网膜的表达变化。方法:采用模拟人类膳食结构的长期高脂肪饮食24wk加小剂量STZ注射复制2型糖尿病大鼠模型8只,并以正常饮食组健康大鼠8只作为对照;应用免疫组织化学和原位杂交技术检测了TNF-α,IL-6及IL-18蛋白及mRNA在2型糖尿病大鼠视网膜的表达变化,并应用病理图像系统对结果进行定量分析。结果:TNF-α,IL-6及IL-18蛋白和mRNA在2型糖尿病组大鼠视网膜中均呈阳性表达;与正常组大鼠相比,其平均灰度、平均吸光度以及积分吸光度均有显著性差异(P<0.05)。结论:TNF-α,IL-6及IL-18在2型DM大鼠视网膜的表达增高。
AIM: To investigate the expression of adipose inflammatory factors (TNF-α IL-6 and IL-18) in retina of type 2 diabetic rats.
METHODS: Eight male SD rats were fed with a high-fat diet for 24 weeks and then were injected with low-dose streptozototin intraperitoneally to replicate rat models of type 2 diabetes mellitus, and eight rats were fed with normal diet as controls. The expressions of TNF-α, IL-6 and IL-18 in retina of diabetic rats were determined by immunohistochemistry and in situ hybridization. The results were analyzed quantitatively by pathologic image software.
RESULTS: Both protein and mRNA content of TNF-α, IL-6 and IL-18 in retina of diabetic rats were higher than those in normal rats. Statistical differences were found signirfiant in average gray (AG), average optical density (AOD) and integral optical density (IOD) (P 〈0.05).
CONCLUSION: High expressions of TNF-α, IL-6 and IL-18 in retina of type 2 diabetic rats indicated that the adipose inflammatory factors might strengthen the inflammatory reaction and increase insulin resistance, and thus be involved in the occurrence of diabetic retinopathy.
