血清标本的保存温度和时间对乙型肝炎两对半定量测定的影响 被引量:10

The influence of serum preservation temperature and time to HBV markers quantitative detection

摘要 目的探讨血清标本保存时间和温度件对乙型肝炎(下称乙肝)两对半定量检测结果的影响,寻求保存血清标本的理想方法。方法分别采用2~8℃和-20℃两种温度保存标本,应用雅培i2000SR化学发光仪定量测定两对半指标,2~8℃的保存标本每隔3d检测1次,-20℃的保存标本按照1、3、5、6个月周期检测。结果2~8℃保存标本7d,结果差异无统计学意义(P>0.05),超过7d,乙肝病毒e抗原和核心抗体结果发生变化,差异有统计学意义(P<0.05);-20℃连续保存6个月,测定结果无统计学意义(P>0.05)。结论标本的保存温度和时间直接影响乙肝两对半定量结果的准确性,在各种保存条件下,随着温度的下降,标本保存时间延长。2~8℃标本可稳定7d;-20℃可以保存6个月。 Objective To discuss the influence of serum specimen retention time and the temperature to HBV markers (HBVM) quota examination result, so as to seek for the ideal method for serum specimen preservation. Methods Two kind of temperatures (2-8 ℃ and -20 ℃) were selected to preserve serum samples. Abbott i2000 chemiluminescence meter was applied to quantitatively determining HBVM. 2-8 ℃preserved specimens were examined once every 3 days; -20 ℃ preserved specimens were examined periodically at 1st month, 3 rd mon, 5 rd month and 6th month. Results There was no significant change of HBVM quantitative detection result during 7 days of preservation at 2-8 ℃. However, HBeAg and anti-HBc changed significantly after the duration (P〈0.05). There was no significant change of HBV-M while continuously preserving specimen for 6 months at -20 ℃ (P〉0. 05). Conclusion The specimen preserved temperature and duration directly affect the accuracy of HBVM quota results. Under all preserved condition, along with the temperature dropping down, the specimen retention time lengthens, stabilizing for 7 day at 2-8 ℃, and -20 ℃ for 6 months.
