高效液相色谱法及高碘酸法测定三苄糖苷的含量 被引量:1

摘要 目的:建立测定三苄糖苷(TBS)两种异构体含量的高效液相色谱法(HPLC)与高碘酸法。方法:采用HPLC法与高碘酸法分别测定了TBS两种异构体的含量,并进行了最佳试验条件的筛选和方法学考察。结果:采用HPLC法测定的TBSα体的含量为99.36%,β体为98.83%;采用高碘酸法测定的TBSα体的含量为99.41%,β体为98.89%。结论:HPLC法灵敏、简便,选择性好,为常用的原料药与制剂的含量测定方法;高碘酸法准确度、精密度均较高,且测定时不需要对照品,主要用于高纯度原料药的含量测定与对照品含量的标定。此结果可为TBS质量控制方法的优化提供参考。  Objective:To establish the high performance liquid chromatography(HPLC)and periodate methods used to detemine the contents of two isomers of tribenoside(TBS).Methods:The contents of two isomers of TBS in the samples were determined by HPLC and periodate method,respectively.The optimal experimental conditions was studied.Results:The content of isomer α was 99.36% and isomer β was 98.83% by HPLC.Meanwhile,the content of isomer α was 99.41% and isomer β was 98.89% by periodate method.Conclusion:HPLC is sensitive and simple,has a good selectivity and can be used to the crude drugs and preparations.Periodate method has good accuracy and precision and can be used to pure crude drugs because this method does not need standard reference substance.This study may be adopted by optimizing method of quality control of TBS.
