肿瘤坏死因子-α与糖尿病心肌病关系的实验研究 被引量:2

摘要 目的通过建立糖尿病模型,观察糖尿病心肌的病理及超微结构改变,并测定心肌肿瘤坏死因子的表达,探讨TNF-α在糖尿病心肌病发生发展中的炎症机制。方法Wistar大鼠实验组用1%的链脲佐菌素溶液腹腔注射制作糖尿病模型,对照组用等量的柠檬酸-柠檬酸钠缓冲液腹腔注射。实验前及注射STZ72h后各组大鼠测体重、尿糖、尾静脉采血测血糖、甘油三酯、总胆固醇、低密度脂蛋白胆固醇、高密度脂蛋白胆固醇。两组标准喂养8w后测体重,快取左心室心肌,制作切片分别行组织学观察心肌病理变化和电镜下观察心肌超微结构。结果实验组糖尿病大鼠光镜下可见明显的心肌纤维排列紊乱,心肌细胞肥大等,且冠状动脉分支无异常改变,电镜下可见心肌肌原纤维稀疏、断裂、走向紊乱,线粒体增多、肿大变性,嵴断裂、稀疏,糖原沉积,间质胶原增生等。结论糖尿病大鼠心肌中TNF-α的表达明显高于对照组,提示TNF-α可能参与了糖尿病心肌病的发生发展。 Objective To observe the pathologoic and ultrastTucture changes of the diabetic myocardium, measure the expression of tumor necrosis factor- α(TNF- α) of myocardium and explore the inflammational mechanism on TNF- α in the development of diabetic cardiomyopathy through the diabetic rat model. Methods Wistar rats were injected with 1% streptozotocin(STZ ) into abdominal cavity to make DM model and the control group were injected with equivalent citromalic acid-sodium citrate buffer. Both groups needed to examine the weight,urine glucose, blood glucose,TG and TC before injection and 72 hours after injection. Both groups were standardly fed for 8 weeks. The left ventricular myocardium was cut to make histological section and electron microscopic section to observe. Results The experimental diabetic rats were observed obviously derangement of cardiac muscle fiber, hypertrophy of cardiac myoscope,meanwhile,observed rarefaction,fragmentation, derangement of myocardium myofibril, proliferation,tumefaction,degeneration of mitochondria, disruption, rarefaction of mitochondrion ridge, deposition of glycogen, hyperplasia of collagen in interstitial under electron microscope, but control rats showed no difference. Conclution The expression of TNF- αin the diabetic rat' s myocardium was obviously higher than that of the control group, which hints the TNF- α might play a role in the development of diabrtic cardiomyopathy.
