摘要
本研究以磺胺二甲嘧啶的单克隆抗体为核心试剂建立ELISA检测方法,可检测鸡饲料和猪饲料中的磺胺二甲嘧啶。ELISA方法的标准曲线为:y=-29.448x+74.565,R2=0.9917,IC50=6.8ng/mL,线性范围为0.7～71.3ng/mL,检测限为0.3ng/mL。选择磷酸盐缓冲溶液(0.01mol/L,pH7.4)为提取溶剂,以0.1、1μg/g和10μg/g的浓度添加于空白的鸡和猪配合饲料时,其回收率为73.3～107.3%,变异系数为0.9%～19.2%。
Based on anti-sulfamethazine monoclonal antibody,an ELISA method was developed to detect sulfamethazine in chicken feed and swine feed.The formula was y=-29.448x＋74,565 ,R2=0.9917,and the 50 % inhibition concentration（ICso）was 6,8 ng/mL with the limit of detection of 0.3 ng/mL.The linear detecting range of calibration curve was from 0.7 ng/mL to 71,3 ng/mL.When the spiked concentrations of sulfamethazine were 0.1μg/g, 1μg/g,and 10μg/g in blank feed ,the recovery was 73.3 % - 107.3 % with coefficients of variation（CV）of 0.9 % - 19.2 %.
