Abnormal expression of adipose-related genes in TAO adipose tissue

摘要 目的探讨甲状腺相关眼病(thyroid-associatedophthalm opathy,TAO)患者眼眶脂肪组织相关基因的表达。方法选用TAO患者(20例)及正常对照(17例)眼眶脂肪组织,采用RT-PCR法对脂肪细胞分化转录因子前脂肪细胞因子-1(Pref-1)、过氧化物酶体增殖体激活受体γ(PPARγ),脂肪细胞分泌因子瘦素、脂联素以及共同抗原TSHR等5个基因进行检测。比较TAO及正常人眶脂肪组织基因表达的改变。结果RT-PCR法测得的TAO患者眼眶脂肪组织中各基因mRNA表达的相对灰度值为:瘦素1.23±0.03、脂联素1.16±0.08、PPARγ1.03±0.06、TSHR0.96±0.04、Pref-10.48±0.07,其中瘦素、脂联素、PPARγ、TSHRmRNA表达明显较正常对照组增强,而Pref-1表达则减弱(P<0.05)。结论TAO患者眼眶脂肪组织中存在着异常的基因表达,在TAO的发生、发展中可能起着重要作用。 Objective To discuss changes of adipose-related genes expression in thyroid-associated ophthalmopathy(TAO) adipose tissue. Methods Adipose tissues were collected from 20 TAO patients and 17 normal control people. Using RT-PCR method to detect the genes expression of adipocellular differentiation and transcription factor ( pref-1 ), PPAR, leptin, adiponectin and TSHR,which were also Compared between the two groups. Results The relative gradations of mRNA expression in TAO orbital tissues are Leptin 1.23 ± 0. 03 ,Adiponectin 1.16 ±0.08,PPARγ 1.03 ±0.06,TSHR 0.96 ±0.04,Pref-1 0.48 ± 0. 07. The expression of PPARγ, leptin, adiponectin and TSHR were upregtdated in TAO adipose tissues compared to controls, while lower expression of pref-1 was also found in TAO patients ( P 〈 0.05 ). Conclusion There are abnormal gene expressions in TAO adipose tissues,and it may make important effects on the development of TAO.
