苦豆子生物碱对内毒素的体外灭活作用 被引量:9

摘要 目的观察苦豆子生物碱对内毒素(LPS)的体外灭活作用。方法利用鲎试剂显色基质法定量测定苦豆子总碱、槐果碱、槐定碱、氧化苦参碱体外灭活LPS的作用及其剂量效应关系。结果槐果碱浓度为1.25μg/ml、槐定碱5μg/ml,作用1小时即有显著灭活LPS的作用,灭活百分率分别为79.04%和79.34%;苦豆子总碱浓度为40μg/ml时灭活LPS达77.20%;氧化苦参碱浓度为125μg/ml时可减少LPS 44.68%。4种苦豆子碱对LPS的灭活率在一定范围内随药物浓度增加而提高。结论苦豆子总碱及3种单体碱具有体外直接灭活内毒素作用,并在一定范围内呈现量效关系。 Objective To study the role of four kinds of Sophora Alopecuroides L. Alkaloids (total alkaloids of sophora alopecuroides, sophocarpine, sophoridine, oxymatrine) deactivating endotoxin in vitro. Methods The role of four kinds of Sophora Alopecuroides L. Alkaloids deactivating endotoxin in vitro was detected by quantitative chromogenic tachypleus amoebocyte lysate. Results Sophocarpine and sophoridine had deactivating endotoxin effect even when they were 1. 25μg/ml, 5μg/ml. The deactivating rate of LPS was 79.04% and 79.34%, respectively. Wtien total alkaloids of sophora alopecuroides was 40μg/ml, it deactivated 77.20% LPS and when oxymatrine was 125μg/ml, the deactivating rate of LPS was 44.68%. The action of Total alkaloids of Sophora Alopecuroides and three kinds of Sophora Alopecuroides L. Alkaloids on deactivating endotoxin in vitro were increased in certain extent as concentrations were heightened. Conclusion The results show that Sophora Alopecuroides L. Alkaloids has straight deactivating endotoxin affect in vitro in a dose -dependent manner.
