摘要
目的：研究肾癌中Ｃ－ｍｙｃ基因的扩增和表达情况及探讨Ｃ－ｍｙｃ基因与肾癌恶性表型及预后的关系。方法：应用核酸杂交技术对４５例肾癌和５例正常肾赃组织的Ｃ－ｍｙｃ基因的扩增进行检测，同时用免疫组化方法检测Ｃ－ｍｙｃ蛋白产物的表达水平。结果，１５例肾癌中有基因扩增，扩增率３３．３％，但与临床分期与病理分级无关；２７例肾癌组织有Ｃ－ｍｙｃ蛋白产物的阳性表达，阳性率为６０％，正常组织和癌旁组织Ｃ－ｍｙｃ蛋白为阴性，并且Ｃ－ｍｙｃ蛋白表达与肿瘤的临床分朝和病理分级有关。结论：Ｃ－ｍｙｃ基因可能是肾癌发生的相关基因之一，Ｃ－ｍｙｃ蛋白表达与肾癌临床行为关系密切，有可能成为肾癌诊断和预后的一个重要分子生物学检测指标。
By using dot bloting hybridization analy- sis , the C-myc gene amplification was detected in 45 cas- es of renal cancers and 5 cases of normal kidneys. C-my. gen amplification was observed in 15 out of 45 renal can- cer. wit h the amplification rate being 33. 3% , It was also found the amplification was not related to the clinically grading and staging. The overexpression of C-cyc gene was determined by immunohistochemical techmique in 45 cases of renal cancers. The results showed that C-myc positive expression was found in 27 cases of renal can- cer. with the positive rate being 60% , while normal tis- sue gave negative result. Overexpression of C-myc was associated with histopathological garde and clinical stage of bladder cancer. The above findings suggest the the C- myc gene may be one of the oncogenes involving renal carcinogenesis . and C-nayc gene expression be related to clinical behar.ious of renal cancer , which may be applica- ble as a tumor marker for evaluation of malignant poten- t ial invasiveness and prognosis of renal cancer.
