摘要
为了更好地利用气相色谱法鉴别一些新来源的菌种如极端环境微生物和具有新的应用价值的微生物,本论文研究了微生物菌体的收集、干燥、甲酯化和气相色谱分析条件.建立了气相色谱分析微生物细胞内脂肪酸的方法,应用该方法分析了As1.643、As1.1130、As1.1807、HWS、DSM.6577菌细胞内的脂肪酸.结果表明,本方法可以分析细胞内16种脂肪酸.在所分析的5种菌中,最多的含有11种脂肪酸,最少的含有7种.通过对其脂肪酸种类的对比可以分辨出这5种菌种,并且得出了这5种菌的指纹图谱,可用于对菌种进行鉴定.
An method of gas chromatography by determining the fatty acids in cells was developed for identification of bacteria. The optimum conditions of separation, collection, methylation and gas chromatography were tested. When this method was applied to detect the fatty acids in cells of As1. 643, As1. 1130, As1. 1807, HWS, DSM. 6577, 16 kinds of fatty acids can be detected. The results showed that different kind of fatty acids occurred in the 5 tested bacteria, which can be used to identifying these bacteria.
