组织型谷氨酰胺转移酶在开角型青光眼患者房角组织的表达

The expression of tissue transglutaminase in human tissue from the anterior chamber angle of eyes of patients with primary open angle glaucoma

摘要 目的探讨组织型谷氨酰胺转移酶(tissue transglu-taminase,tTG)在原发性开角型青光眼患者房角组织中的表达情况及其与闭角型青光眼患者和正常人眼房角组织中表达情况的比较。方法术中取青光眼患者房角组织,采用免疫组织化学染色方法观察开角型、闭角型青光眼患者及正常人房角组织内tTG的表达情况,用RT-PCR方法分析组织内tTG的表达水平。结果免疫组织化学及RT-PCR结果均显示,正常组与闭角型青光眼组房角tTG的表达差异并无显著性(分别为0.180±0.032,0.212±0.019,P>0.05),而正常组与开角型青光眼组、闭角型青光眼组与开角型青光眼组房角组织tTG的表达差异有极显著性(分别为0.180±0.032,0.325±0.052;0.212±0.019,0.325±0.052,P<0.01)。结论开角型青光眼房角组织tTG的表达增加,tTG的增加可能与开角型青光眼的发病有关。 Objective To study the expression of tissue transglutaminase (tTG) in the chamber angle of patients who have been diagnosed with primary open angle glaucoma (POAG) and to compare the chamber angle with patients who have been diagnosed with primary angle closure glaucoma (PACG) and with normal people. Methods Human tissue from anterior chamber angles was obtained 1 hour after trabeculectomys and was fixed in 4% paraformaldehyde for 3 hours. Then the tissue were cut into sections. In all, there were 26 POAG and 17 PACG eyes. In addition, we obtained 3 normal samples from corneal transplant donors. The tissue was divided into 3 groups: group 1 was the normal group, group 2 was the PACG group, and group 3 was the POAG group. The expression of tissue transglutaminase protein was detected by an immunohistochemical technique with antibodies against tTG. The sections were observed under light microscopy and scanned. Using an image analysis system for calculation, the optical density value (OD value) was obtained, SPSS was used to calculate OD values, In the second step, tissue was analyzed for tTG using reverse transcription polymerase chain reaction (RT-PCR), Tissue from the different groups was then placed in a homogenizer and a Trizol kit was used to extract RNA, A tTG primer was added to transform RNA to cDNA, The samples were then passed through electrophoresis so expression could be observed, Results The results of immunohistochemistry showed that tTG was expressed much more in the POAG group than in the other groups. Photodensitometry showed that there was a significant difference between the POAG group and the normal group, (P 〈0.01,POAG: 0.325±0.052 and normal: 0.180±0.032), and there was a significant difference between the POAG group and the PACG group, (P〈0.05,POAG: 0.325±0.052 and PACG: 0.212±0.019).
