摘要
利用胃蛋白酶消化小鼠肌肉,获取了旋毛虫种本地毛形线虫(T.nativa)的肌幼虫虫体。利用TRIZOL混合虫体直接提取肌幼虫的总RNA,通过分光光度计Ultrospec 3000检测结果,以及用RT-PCR方法扩增出了编码T.na- tiva 49 kDa的ES蛋白结构基因,后续试验结果显示改进的一步法提取本地毛形线虫总RNA效果很理想。
The muscle--stage larvae of Trichinella nativa (T. nativa) were collected digested by pepsin- HC1. The total RNA of T. nativa was isolated using TRIZOL according to the manufacture's instruction after the worms being pestled together with TRIZOL. It was showed that from the results of RNA detection with Ultrospec 3000, and also from the next result that the gene of the worm which encoding 49kDa ES protein of T. nativa was amplified by RT--PCR, that the improved single--step method of RNA isolation for T. nativa we used was ideal.
