摘要
目的建立测定Beagle犬血浆中齐墩果酸的HPLC-MS测定法,以进行齐墩果酸滴丸的生物利用度和药动学研究。方法采用HPLC-MS,以Hypersil GOLD(2.1 mm×150 mm,5μm)色谱柱,流动相为20 mmoL.L-1醋酸铵-乙腈(24∶76),流速0.3 mL.min-1,检测波长为210 nm。结果Beagle犬血浆内源性物质不干扰样品的测定,血浆中齐墩果酸浓度在3.88～240.00μg.L-1内线性关系良好(r=0.999 3);最低检测浓度为3.88μg.L-1;方法平均回收率大于85%;平均提取回收率大于80%;日内和日间RSD均小于10%。结论所建立的测定方法准确可靠,符合生物样品分析要求。
OBJECTIVE To establish a method for the determination of oleanolic acid in plasma of Beagle. METHODS The concentrations of oleanolic acid in plasma of Beagle were determined by HPLC-MS. The separation was achieved using a Hypersil GOLD column （ 2. 1 mm×150 mm,5μm） with a mobile phase consisting of ammonium acetate （ 20 mmol·L^-1 ） -acetonitrile （ 24 ： 76 ）. The flow-rate of the mobile phase was set at 0. 3 mL·min^-1, the determinated wavelength was 210 nm. RESULTS The linear calibration curve（r =0. 999 3） was obtained in the concentration range of 3.88 -240. 0 μg ·L^-1. The inter-day and intra-day precision was less than 10%, The average recovery was more than 85%, The extraction efficiency was more than 80%. CONCLUSION The HPLC assay for oleanolic acid measurement in plasma is simple, sensitive and accurate.
