摘要
目的探讨生存素(survivin)拼接体在膀胱移行细胞癌的表达情况及其与肿瘤分期、分级的关系。方法30例膀胱移行细胞癌患者中,Ta/T1期17例,T2~T4期13例;G1/G2级23例,G3级7例;配对的癌旁组织8例;另有8例正常膀胱黏膜取自非肿瘤患者。采用TRIzol一步法提取总RNA,逆转录合成cDNA,再进行PCR扩增。所有标本用GAPDH mRNA作内对照,目的PCR条带用BIO-RAD Quantity One4.5软件根据面积与吸光度定量分析。结果30例膀胱移行细胞癌标本有28例检测到生存素及其拼接变异体(生存素-△Ex3和生存素-2B) mRNA的表达,阳性率为93.3%。生存素为主要的转录本(0.712±0.120),生存素-△Ex3(0.101±0.029)和生存素-2B(0.214±0.076) mRNA为低水平表达,三者的表达水平有显著性差异(P<0.01)。与正常膀胱黏膜和癌旁组织相比,膀胱癌组织生存素及其拼接变异体mRNA的表达水平均明显增高(P<0.01)。而生存素-2B mRNA表达水平在浸润性、高级别癌明显低于非浸润性、低级别癌(P<0.01)。结论生存素拼接变异体可表达于膀胱癌组织,且生存素-2B在浸润性、高级别膀胱癌中表达降低,推测其抗凋亡活性较生存素和生存素-△Ex3为低。
Objective To investigate the expressions of survivin and its variants in bladder transitional cell carcinoma and the relationship between the expression and the stage or grade of bladder cancer. Methods Of 30 patients with bladder transitional cell carcinoma, 17 cases were at stage of Ta or T1, 13 cases T2 -T4. Twenty three cases were in degree of G1 / G2 and 7 cases G3. Eight cases had the matched pairs of non-neoplastic tissue samples. Eight samples of normal bladder mucosa were from non-tumor cases. Total RNA was extracted from bladder cancer tissues according to the TRIzol protocol, cDNA was synthesized from total RNA of each sample, and cDNA was used for PCR. Glyceraldehyde phosphate dehydrogenase(GAPDH) was also amplified as an internal control. The PCR bands were analyzed by BIO-RAD Quantity One 4.5 software. Results In 28 cases of 30 tumor tissue samples, the expressions of survivin and its variants were detected. Survivin was the predominant transcript variant, whereas only low level of survivin-△Ex3 and survivin-2B were observed. The differences of expression between the survivin variants were statistically significant ( P 〈 0. 01). In contrast, a significant decrease in the expression of the non-anti-apoptotic survivin-2B variant became evident at advanced stages or grades of bladder cancer (P〈 0. 01 ). Conclusion We have found the variants of survivin in bladder cancer, and demonstrated that the expression of survivin-2B at high stage or grade bladder cancer is down-regulated, indicating that its antiapoptotic function may be lower than other variants.
