摘要
用电化学和光谱学方法研究了Al(Ⅲ)与DA配合物的形成,测定了配合物的配比和配位常数。在pH4.4,7.0,8.5条件下,Al(Ⅲ)与DA的配比分别为1∶1,1∶2,1∶3;DA与Al(Ⅲ)的配位数分别为2.29×1011,1.13×1011,3.17×1011L2/mol2。分别用电化学和光谱法研究了Al(Ⅲ)和DA配合物与DNA的相互作用,确定了作用方式为嵌入式,并测定了配合物与DNA的结合数和结合常数,结合数m=2,结合常数β=1.78×108,表明配合物与DNA发生了较强的相互作用。同时利用Al(Ⅲ)和DA配合物与DNA相互作用后能使体系的氧化峰电流减小,并且体系的氧化峰电流的降低与DNA的浓度成线性关系,其线性方程△Ⅰ(μA)=12.0053cDNA(10-3mol/L)+0.8962,相关系数r=0.9976,以此为基础上提出了DNA的电化学测定方法。
The formation of Al( Ⅲ ) complex with DA was investigated by electrochemistry and spectroscopy. The complex ratio and complex constant were determined in this paper. The complex ratio are 1 : 1,1 : 2 and 1 : 3, and the complex constant are 2.29 × 10^11, 1.13 ×10^11 and 3. 17 ^ 10^11 L^2/mol^2, at pH 4.4,70,8.5, respectively. The interaction of the complex with DNA was studied by electrochemistry and spectroscopy. The results showed that the binding mode is partial intercalation. The complex ratio and complex stability constant of the complex with DNA are 2 and 1.78 × 10^8 ,respectively. Oxidation peak current of complex Al( Ⅲ ) -( DA)2 decreased with the addition of DNA. The decreasing in peak current was proportional to the concentration of DNA. The decreasing in peak current was proportional to the comcentration of DNA. The linear fitting equation is △I(μA) = 12. 0053 cDNA (10^-3mol/L) + 0. 8962 ,the correlation coefficient is 0. 9976. This proposed method can be used in DNA determination.
