大鼠肾上腺交感神经离断对肾上腺髓质增生的治疗作用

Experimental study of denervation of adrenal sympathetic nerve in the treatment of rat adrenal medulla hyperplasta

摘要 目的观察大鼠肾上腺交感神经离断对于肾上腺髓质增生的治疗作用。方法雄性Wistar大鼠80只随机平均分为A、B、C、D四组。A组为单侧(左侧)肾上腺交感神经离断组,B组为假手术组,C组为肾上腺髓质增生组,D组为正常对照组。A、B组术后1周同C组给予相同剂量利血平6周。C组停药后1周测定激素水平并处死5只大鼠,测定肾上腺髓质百分数。剩下大鼠行双侧肾上腺交感神经离断,术后1个月处死并与1个月前比较。结果A、B组激素水平明显高于D组,A组手术侧(左侧)肾上腺髓质百分数明显低于未手术侧(右侧,P<0.05),而与D组无明显差异。C组双侧肾上腺交感神经离断术的大鼠,术后激素水平较术前有所下降。结论大鼠肾上腺交感神经离断可抑制利血平诱导大鼠肾上腺髓质增生,同时降低髓质增生大鼠的激素水平。 Objective To study the effect of denervation of adrenal sympathetic nerve in the treatment of rat adrenal medulla hyperplasia induced by reserpine. Methods Eighty male Wistar rats were randomly divided into 4 groups : group A, unilateral adrenal denervation ( the left) ; group B, pseudo-operation; group C, experimental model of adrenal medulla hyperplasia; and group D, normal control. The rats in group C were subcutaneously injected with reserpine (0.4 mg/kg every day) and 0.9% sodium chloride (0.4 mg/kg every day). One week after surgery,the rats in groups A and B were injected with reserpine just as group C for 6 weeks. The rats in group D were subcutaneously injected with 0.9% sodium chloride (0.4 mg/kg-every day). Catecholamine (CA) level in plasma and 3-methoxy-4-hydroxyvanillymandeic acid (VMA) content in the urine were measured at first week after injection in group C and pathologic studies of adrenal were also done. The rest rats in group C were subjected to bilateral denervation and killed one month after operation. The biochemical index and pathological changes were measured. Results At different time points,the values of VMA and CA in groups A and B were higher than those in group D. The percent of adrenal medullas in the left adrenals of group A was lower than the right (P 〈 0.05). The values of VMA and CA in group C postoperation were lower than those preoperation. Conclusion Denervation of adrenal sympathetic nerve can restrain the proliferation of adrenal medulla induced by reserpin, and at the same time it can decrease the hormone level of adrenal medulla hyperplasia.
