止喘胶囊对哮喘大鼠气道重建转化生长因子β1和血小板源性生长因子蛋白及其mRNA表达的影响 被引量:1

摘要 目的探讨止喘胶囊对哮喘大鼠肺组织TGF-β1、PDGF-B蛋白及其mRNA表达的影响。方法将48只雄性SD大鼠随机分为正常对照组、哮喘模型组、止喘胶囊组、地塞米松组、止喘胶囊加地塞米松组五组,每组8只。通过腹腔注射卵蛋白、右腿肌注氢氧化铝致敏及反复雾化吸入不同浓度(1%、2%、3%)卵蛋白溶液建立哮喘气道重建动物模型。在哮喘激发4周后检测肺组织TGF-β1、PDGF-B蛋白及其mRNA表达。计量资料用ANOVA进行分析。结果哮喘模型组大鼠气道上皮TGF-β1、PDGF-B蛋白以及mRNA表达显著高于正常对照组大鼠(P<0.01);止喘胶囊组、止喘胶囊加地塞米松组和地塞米松组大鼠TGF-β1、TGF-β1mRNA表达显著低于哮喘模型组大鼠(P<0.01或P<0.05)。结论止喘胶囊通过减少气道上皮TGF-β1、PDGF-BmRNA表达从而减轻气道炎症及气道重建,降低气道高反应性,从而发挥固本防喘作用。 Objective:To evaluate the effects of "Zhichuan Capsule" on expressions of TGF-β1,PDGF-B in airway remodeling of bronchial asthmatic rats. Methods:The model was established by different density ovalbumin (1%,2%,3%)sensitized and activated in prolonged and repeated exposure. The rats were randomly divided into normal control,model group, "Zhichuan Capsule" group, dexamethasone group,and "Zhichuan Capsule"plus dexamethasone group. General histological changes were observed by hematoxylin and eosin stained sections after 4- week activation. The expression of TGF-β1,TGF-β1 mRNA,and the airway was examined by immunocytochemistry and RT-PCR. Statistical comparison was performed by ANOVA followed by Fisher LSD test. Results:The expressions of TGF-β1,PDGF-B protein and mRNA, TGF-β1 mRNA in model group were higher than those in normal control group (P〈0.05). The expressions of TGF-β1,TGF-β1 protein and mRNA in "Zhichuan Capsule group","Zhichuan Capsule" plus dexamethasone group,and dexamethasone group were lower than those in model group (P〈 0.01 or P〈 0.05). Conclusion: "Zhichuan Capsule" can down-regulate the expressions of PDGF-B,TGF-β1 protein and mRNA,and suppress airway remodeling.
