西兰花叶叶绿素提取及叶绿素铜钠盐制备工艺研究 被引量:20

Study on the Extraction Process of Chlorophyllin from Brassica Leaves and the Preparation of Sodium Copper Chlorophyllin from Its Leaves

摘要 研究了以西兰花叶为原料提取叶绿素制备叶绿素铜钠盐的工艺条件。通过正交试验对西兰花叶叶绿素提取工艺进行了优化,结果表明西兰花叶叶绿素提取的最佳条件为:乙醇浓度95%;料液比1∶6;萃取温度为70℃;萃取时间2 h;叶绿素铜钠盐制备条件为:5%NaOH,65℃皂化30 min;1%硫酸调pH 2.0,65℃酸化30 min;15%硫酸铜,65℃铜代反应60 min,成盐NaOH调pH 12,65℃1 h。 The paper is about the preparation of sodium copper chlorophyllin from Brassica oleacea leaves. The extraction process was optimized by orthogonal experimental design. Experimental results show that the optimal extracting conditions were obtained as follows: the solvent is 95% ethanol, the ratio of solute to solvent is 1 : 6, the temperature is 80℃ and the extracting time is 2 hours. The conditions of the preparation of sodium copper chlorophyllin are following: saponification with 5% NaOH for at 65 ℃ 30 min, acidifying treatment by H2SO4 at pH 2.1 65℃for 30 min, reaction with 15%CuSO4 at 65 ℃ for 30 min, copper chlorophyll by 2% NaOH at 65℃ forl hour.
