离子交换法从发酵液中分离提取谷氨酰胺的试验研究

摘要 本文介绍了采用D330做离子交换材料提取谷氨酰胺的机理,并从pH、柱高、柱径和经过处理的树脂对谷氨酸上柱的影响展开实验研究,并得出相应结论。D330型树脂其吸附容量在pH3.2时达最大,并随高径的增加而成倍增加,但此时树脂利用率较低只达到40%左右,而谷胺酰氨的收率在70%左右。未能达到完全的分离回收效果。因此对离子交换柱进行工艺改进,采用2A-2B双柱串连的工艺,2A柱采用的是D151弱酸阳树脂柱,2B采用弱碱D330阴树脂,经过堆2A-2B上柱试验研究,树脂利用率达到80%以上,谷氨酸去除率达到100%,谷胺酰氨收率可以达到91.8%。 The mechanism of separating Glutamine with D330 resin was introduced in this paper. The experiment was based on studying the different pH and different height and diameter on the effect of the adsorption of glutamic acid. Adsorption capability reaches the most at pH 3.2 and increases by the increase of height and di- ameter. But only 40% resin was used. The collection rate of Glutamine reached to 70% and couldnt reach completely separation. So the ion exchange colume was changed. The series of 2A -2B was applied,2A was made of D151 resin and 2B was made of D330 resin. By experimenting, 80% resin was used and the removal rate of glutamic acid was 100% ,and the collection of the Glutamine was 91.8%.
