摘要
目的观察茶多酚对脑缺血大鼠脑内少突胶质细胞(OLG)的影响。方法将雄性SD大鼠随机分为假手术组、对照组、茶多酚组。制备全脑缺血模型后,茶多酚组大鼠被分为3组分别按剂量25mg/kg、50mg/kg、100mg/kg茶多酚予以腹腔注射,每日1次,连续7d;免疫组化法检测各组大鼠皮质前体OLG、未成熟OLG及成熟OLG的变化。结果与假手术组比较,对照组皮质前体OLG明显增多,未成熟OLG及成熟OLG明显减少(均P<0.05);与对照组比较,茶多酚干预后皮质前体OLG有不同程度的减少,未成熟OLG及成熟OLG则有不同程度的增加,差异有统计学意义(25mg/kg、100mg/kg,均P<0.05;50mg/kg,P<0.01)。结论茶多酚对OLG缺血性反应有保护作用,并呈现剂量依赖性。
Objective To investigate the effects of tea polyphenol on oligodendrocyte (OLG) in the cerebral ischemic rats. Methods Male Sprague Dawley rats were randomly divided into sham-operated group, control group and tea polyphenol group. The models of whole-brain ischemia were established . Then the rats of tea pnlyphenol group were divided another 3 groups and injected intraperitoneally with tea polyphenol at diffenrent dose (25 mg/kg, 50 mg/kg, 100 mg/kg, respectively) once a day for seven days . The expressions of precursor OLG cells, pedo-OLG cells and mature OLG cells in the cortex were examined by immunohistochemistry and the positive cells were counted. Results Compared with sham-operated group, the number of precursor OLG cells increased and the number of pedo- OLG cells and mature OLG ceils decreased significantly in the control group (all P 〈0.05 ). Compared with control group, the number of precursor OLG cells decreased significantly after tea polyphenol treatment, while the number pedo-OLG cells and mature OLG cells increased significantly(25 mg/kg, 100 mg/kg ,all P 〈0.05; 50 mg/kg, P 〈 0.01 ). Conclusion Tea polyphenol played a protective role in OLG ischemic reaction at a dose dependent manner.
