天然药物槟榔碱对氧化低密度脂蛋白致血管内皮细胞损伤的保护作用研究 被引量:15

摘要 研究天然药物槟榔碱对培养的牛主动脉内皮细胞氧化损伤时MCP-1和ICAM-1mRNA表达水平的影响,探讨其抗动脉粥样硬化的作用机制。用血管内皮细胞与不同浓度槟榔碱预孵育20h后,给予氧化低密度脂蛋白损伤,提取各组细胞总RNA,通过逆转录-聚合酶链式反应技术,以β2-微球蛋白为内参照,检测内皮细胞MCP-1和ICAM-1mRNA表达水平的变化。得到内皮细胞与槟榔碱预孵育后,其MCP-1和ICAM-1mRNA的表达水平明显低于与氧化低密度脂蛋白直接接触的细胞。说明氧化低密度脂蛋白可诱导牛主动脉内皮细胞MCP-1和ICAM-1mRNA表达水平增加,槟榔碱可以减轻其过量表达,保护内皮细胞。 abstract:To investigate the effectsof arecoline against MCP-1 and ICAM-1 mRNA expression in cultured endothelial cells derived from bovine aorta,the ICAM-1 and MCP-1 mRNA expression in bovine aorta endothelial cells was measured by reverse transcription-polymerase chain reaction and normalized to the mRNA level of /32-microglobulin in the absence or in the presence of ox-LDL or land pivanampeta. It is resulted that both MCP- 1 and ICAM-1 mRNA expression in the endothelial cells derived from bovine aorta were increased by ox-LDL stimulation ,which could be prevented by arecoline at the concentrations of 10 and 100μmol/L. It is conclused that the over expressionof MCP-1 and ICAM-1 in endothelial cells induced by ox-LDL could be blocked by arecoline. Arecoline can protect the endothelial cellsfrom the oxidative injury.
