酸水解法提取甘草中异甘草素工艺研究 被引量:5

摘要 目的以甘草粉末为原料,研究了酸水解法提取异甘草素的工艺。方法采用正交实验考察盐酸浓度、水解时间、水解温度、料液比(甘草重量:盐酸体积)4个因素对异甘草素提取率的影响,确定酸水解法提取异甘草素的最佳工艺。结果酸水解法提取异甘草素的最佳工艺参数为盐酸浓度1mol·L-1,水解作用时间2h,水解温度90℃,料液比1∶5。水解后碱中和pH为7,过滤后用体积分数80%乙醇提取滤渣,料液比1∶10,超声20min,提取2次;乙酸乙酯萃取滤液,液液比1∶1,萃取2次。结论酸水解法提取异甘草素的提取率为2.47‰,浸膏中异甘草素含量为3.01%,其提取率和含量分别是索氏提取的8.82和10.03倍,是乙醇超声提取的9.88和9.41倍。 OBJECTIVE To study the extraction process of isoliquiritigenin from licorice by acid hydrolysis. METHODS Four extraction factors influencing the extraction yield of isoliquiritigenin were studied, such as concentration of hydrochloric acid ( HCl), acid hydrolysis temperature,acid hydrolysis time and the ratio of material to liquid (weight of licorice:volume of HCl). RESULTS The orthogonal test indicated the optimum method of acid hydrolysis extraction conditions of isoliquiritigenin were as follows : 1 mol · L^- 1 HCl, the ratio of material to liquid of 1 : 5, acid hydrolysis at 90 ℃ for 2 h. The products of acid hydrolysis were adjusted to pH = 7, then filtrated. The residues were extracted by 80% alcohol according to the ratio of material to liquid of 1: 10, ultrasonic extraction for 20 min and extracted two times. Solvent extraction was accomplished by addition of EtOAc to the filtrated sample ( 1 : 1 ) followed by two times. CONCLUSION The extraction yield of isoliquiritigenin by acid hydrolysis method is 2.47‰ and the isoliquiritigenin content of ointment is 3. 01%. The extraction yield and the content of isoliquiritigenin are 8.82 times and 10. 03 times as high as those of Soxhlet extraction and are 9. 88 times and 9.41 times as high as those of ultrasonic extraction by 80% alcohol.
