五氯酚钠急性染毒对小鼠免疫细胞损伤影响 被引量:6

Effect of pentachlorophenol sodium on acute damage of splenocyte during acute exposure

摘要 目的观察五氯酚钠对小鼠细胞免疫、体液免疫和巨噬细胞功能的毒性作用。方法将五氯酚钠以25,50和100mg/kg经灌胃方式一次给予小鼠,然后观察给药后3,24和48h小鼠各项免疫功能的变化,采用T淋巴细胞增殖法测定细胞免疫功能,采用B淋巴细胞增殖法测定体液免疫功能,采用巨噬细胞吞噬能力和分泌NO的能力来测定巨噬细胞功能。结果100mg/kg五氯酚钠染毒后3h,小鼠的脾脏和胸腺相对重量较对照组降低,脾脏B淋巴细胞的增殖能力受到抑制,腹腔巨噬细胞的吞噬功能和NO生成能力受到抑制。25和50mg/kg五氯酚钠在染毒后24 h依然对脾细胞中B淋巴细胞增殖能力产生抑制作用。这种抑制效应到48h时完全消失。结论五氯酚钠一次染毒在25~100 mg/kg剂量范围内对小鼠体液免疫和细胞免疫功能产生抑制作用,100mg/kg对巨噬细胞功能产生抑制作用,对细胞免疫功能未见影响。 Objective To observe the effect of pentachlorophenot sodium on cellular immunity, humoral immunity and macrophage function of mice. Methods Pentachlorophenol sodium was garaged to mice once time and the dosages of pentachlorophenol sodium were 25, 50 and 100 mg/kg. The changes of immume function were observed at 3,24 and 48 hours after pentachlorophenol sodium treatment. Cellular immunity was measured through T lymphocyte proliferation; humoral immunity was measured through B lymphocyte proliferation; Macrophage function was measured through phagocytic function and NO production. Results 10 mg/kg pentachlorophenol sodium could decrease the weight and relative weight of spleen and thymus when compared with control mice at 3 hours after treatment. At the same time B lymphocyte proliferation, phagocytic function and NO production were also decreased. 25 and 50 mg/kg pentachlorophenol sodium reduced the B lymphocyte proliferation not only at the hours after treatment but also the 24 hours after treatment. At 48 hours after treatment there were no difference about all related index between pentachlorophenol sodium treated mice and control mice. Conclusion 25 to 100 mg/kg pentachlorophenol sodium could inhibit the humoral immunity and 100 mg/kg could inhibit the macrophage function after once treatment. Whereas the cellular immunity was not influenced by current treatment.
